Abstract

The reduction in the number of rare and endangered plants requires the development and implementation of alternative ways of reproduction, including in vitro culture technology. Ornamental groups of plants are of particular importance. Lilium pilosiusculum (Freyn) Miscz is one of the wild growing ornamental species of plants in Yakutia. This rare plant species of «2 b» category is undergoing a decline in numbers due to deforestation, collection, digging of bulbs and requires special protection precautions. This study optimised the conditions under which microclones of Lilium pilosiusculum were obtained. The leaf and bulb explants obtained from a wild plant growing on the territory of the Nyurba ulus of Yakutia were used for regeneration. The most optimal for obtaining sprouts in vitro culture is Murashige and Skoog medium (MS) with the addition of benzylaminopurine (BAP, 1 mg/L) and 2,4-dichlorophenoxyacetic acid (0,5 mg/L). Initiation of sprouting on the basis of bulb scales was achieved during cultivation under conditions of plant shelf with grow lights on MS medium with the addition of BAP at a concentration of 0.5 mg/L. Obtained microshoots of Lilium pilosiusculum were separated and transplanted for rhizogenesis initiation and for studying the adaptation of regenerants in the conditions of various soil substrates.
