Abstract
The effect of different composition of nutrient media, type of explants (hypocotyls, epicotyls, seedlings, leaf fragments) on callusogenesis of two varieties and hybrids of Cucumis sativus L. and four varieties and hybrids of Beta vulgaris L. The sterilization mode of seeds was selected, which were further germinated under aseptic conditions. Different types of explants were obtained from the obtained seedlings and used to obtain callus tissues. The explants were planted on modified Murashige-Skoog (MS)-based media containing various growth stimulants. The optimal medium for obtaining callus tissue from cucumber varieties was MS medium containing 2 mg/L 2,4-dichlorophenoxyacetic acid, while providing a light regime with a 16-hour photoperiod and light intensity of 2200 lux perm2, at a temperature of 20 to 24 ºC. The most successful use of epicotyls of the variety 'Ochiai 9' as explants. The best medium for the formation of callus tissue was MS medium with the addition of 0.1 mg/l 2,4-dichlorophenoxyacetic acid and 1 mg/l 6-benzylaminopurine for the studied varieties 'Pomegranate Ball', 'Gypsy Baron', 'Kombat', 'Sankina Lubov' and hybrid 'Partner F1', the best type of explant - fragments of leaf plate. The callus obtained as a result of the work can be used to study the regeneration ability, used to obtain new varieties by methods of accelerated selection.
